cDNA-derived RNA phage assembly reveals critical residues in the maturation protein of the Pseudomonas aeruginosa leviphage, PP7

PP7 is a leviphage with a single strand of the RNA genome, which infects Pseudomonas Aeruginosa PAO1. A reverse genetic system for PP7 was previously created using Reverse-Transcription CDNA (PP7O) from Virion which was revealed by the Genome RNA. Here, we have found that PP7O CDNA contains 20 nucleotide differences from the sequence of PP7 genomes stored in the database. We create other genetic systems otherwise exploit chemical synthesized CDNA (PP7S) based on the order of databases.

Unlike the PP7O given by transmitted Virion PP7, the PP7S which was revealed by particles was not able to form the plaque in PAO1 cells, which was restored to PAO1 cells expressing maturation protein (MP) from PP7o using this inverse genetic system, we revealed two amino acid residues involved In the known role of the MP (i.e. adsorption and genome replication), it happens to provide a lesson that the RNA virus genome sequencing requires functional verification possible by the genetic significance of the System.

Identification of New Ocellatin Antimicrobial Peptides by CDNA Precursor Cloning in this Frame Family Attractive Peptides

Ocellatins is an antimicrobial peptide family found exclusively in the genus Leptodactylus. Until now, 10 species have been studied and more than 23 peptides are explained. Here we report the number of five new peptides of the Leptodactylus Latrans frog skin (Anura: Leptodactylidae) determined by CDNA cloning from the complete prepro-peptide structure.

Mature peptides are marked in silico tools and compared to those who were previously explained. With 21 amino acid residues, this new set of peptides was previously not described in the Leptodactylus genus section between 100 and 76.2% in common with ocellatin antimicrobial peptides. The new peptides that are cationic and three-dimensional structures (3D) they hold very eternal G1 residues, D4, K7, and K11 and amphipathic high theoretical α-Helix content. Furthermore, in silico analysis of new peptides predicted antimicrobial activity. This research was framed in the previous job context published about Ocellatins, and therefore, providing a review of this family withdrawing peptides.

Construction of cDNA expression libraries in the binary vector using nicking enzyme

Ligation-independent cloning (LIC), such as the Gibson Assembly, tends to produce clones without an insert, depending on the sequence present on the end of the linearized vector. We used the method of enzyme-mediated nicking LIC (NE-LIC) to construct a cDNA library in binary vector pER8. Before building a cDNA library, a pilot experiment conducted, in which GUS coding sequence was cloned into pER8 using NE-LIC.

Approximately 12% of the input vector DNA converted into plasmid carrying the insert GUS, and no plasmid without an insert were detected, indicating that this strategy is very effective for cloning with the binary vector pER8. Therefore, NE-LIC adopted to construct a cDNA library in pER8, using the PCR-amplified cDNA from a library constructed in other vectors. As a result, a cDNA library in pER8 successfully constructed. During the construction of the library, it is important to exclude a plasmid without an insert, because of contamination of the plasmids without inserts decreases the efficiency of the filtering. Therefore, NE-LIC useful for cDNA library construction.

Grapevine berry necrosis in virus (GINV) belongs to the genus in the family Trichovirus Betaflexiviridae. The LN_BETA_RS GINV isolates obtained from the “Beta” grapevine (Vitis riparia × Vitis labrusca) showing chlorotic spots and rings in Xingcheng, Liaoning Province, China. To verify the correlation between GINV and grapevine chlorosis spots and ring spot disease, we constructed an infectious cDNA clone of GINV LN_BETA_RS isolates using a seamless approach to the assembly.
